Understand the basics principles behind the Nelson's assay

Understand what factors influence enzyme-catalysed reactions

Measure the initial rate of the enzyme amylase, at pH 7 and 250C, using starch as a substrate, and manipulate the relevant data in order to draw appropriate graphs.

Measure the Km and Vmax of the enzyme amylase, at pH 7 and 250C, using starch as a substrate, and manipulate the relevant data in order to draw the appropriate graphs.

Understand the relevance of Km and Vmax

Using data provided, measure the effect that amylase concentration has on maltose production at pH 7 and 250C and calculate the specific activity of amylase at pH 7 and 250C

Understand the concept of specific enzyme activity and its relevance in measuring enzyme purification

Be able to perform simple calculations of enzyme activity and specific enzyme activity
